ABSTRACT Local control and overall survival in patients with advanced head and neck squamous cell cancer (HNSCC) remains dismal. Signaling through the Hedgehog (Hh) pathway is associated with epithelial-to-mesenchymal transition (EMT) and activation of the Hh effector transcription factor Gli1 is a poor prognostic factor in this disease setting. Here we report that increased GLI1 expression in the leading edge of HNSCC tumors is further increased by irradition where it contributes to therapeutic inhibition. Hh pathway blockade with cyclopamine suppressed GLI1 activation and enhanced tumor sensitivity to radiotherapy. Further, radiotherapy-induced GLI1 expression was mediated in part by the mTOR/S6K1 pathway. Stroma exposed to radiotherapy promoted rapid tumor repopulation and this effect was suppressed by Hh inhibiiton. Our results demonstrate that Gli1 that is upregulated at the tumor-stroma intersection in HNSCC is elevated by radiotherapy where it contributes to stromal-mediated resistance, and that Hh inhibitors offer a rational strategy to reverse this process to sensitize HNSCC to radiotherapy. Expression of hedgehog signaling molecules as a prognostic indicator of oral squamous cell carcinoma.

BACKGROUND Recent studies have indicated hedgehog pathway plays a role in carcinogenesis of certain cancers. We investigated the clinical significance of its signaling components, including Sonic hedgehog (Shh), Patched (Ptch), and Gli-1, in oral squamous cell carcinoma (OSCC). METHODS By immunohistochemistry, we determined Shh, Ptch, and Gli-1 expression in surgical specimens from 40 patient...

The role of the desert hedgehog signaling pathway during degeneration and regeneration of peripheral nerves

The desert hedgehog (Dhh) signaling pathway is involved in the development of peripheral nerves (PNs). Dhh-null mice show abnormal neuronal development and perineurial barrier function. As it was previously shown that dhh is mainly expressed in developmental nerves and Sonic hedgehog protein (dhh homologous) has therapeutic effects in neuronal survival, we attempted to investigate the possible ...

بررسی جهش‌های رایج ژن‌های KRAS و BRAF در بیماران مبتلا به کارسینومای بافت سنگ‌فرشی سر و گردن

Background: Head and neck squamous cell carcinoma (HNSCC) is the malignancy of squamous cells (the epidermal layer of skin) in cavities in head and neck includes: larynx, pharynx, paranasal sinuses and oral cavity. The main goal of this research was to understand the effect of mutations in two important genes (KRAS and BRAF) in RAS/MAP kinase (EGFR) signaling pathway in tumor cells with head an...
